DESCRIPTION:David Parichy\, Pratt-Ivy Foundation Distinguished Professor of MorphogenesisUniversity of Virginia \n\n\n\n“Pattern formation and cell type diversification in fish pigmentation”Fishes display some of the most striking pigment patterns among vertebrates. These patterns reflect the numbers and arrangements of several classes of neural crest-derived pigment cells\, or chromatophores\, including black melanophores (homologous to mammalian melanocytes)\, yellow-orange xanthophores\, and iridescent iridophores. This talk will explore mechanisms that help organize these cells into patterns\, and how those mechanisms may have been modified to produce different patterns across species. It will also consider how the developmental and evolutionary elaboration of chromatophore classes\, sometimes through terminal additions to differentiation programs or through naturally occurring transdifferentiation\, can contribute to species-specific patterns with behavioral consequences. Together\, these studies suggest that pigment patterns offer a valuable system for understanding the evolution of adult form and for investigating how new cell types arise\, with implications across levels of biological organization.

DESCRIPTION:David Morgan\,  Associate Investigator\, Department of Molecular Integrative PhysiologyUniversity of California\, San Francisco \n\n\n\n“Biochemical Mechanisms of Chromosome Segregation“We study the mechanisms that govern progression through mitosis\, with an emphasis on the complex docking interactions that mediate specific substrate binding to cell cycle enzymes. These efforts have focused recently on separase\, the protease that initiates chromosome separation in anaphase by cleaving a subunit of the cohesin complex. In collaboration with the structural biologist Andreas Boland\, we studied how separase interacts with short sequence motifs on its cohesin substrate\, and we explored the mechanisms underlying the control of separase by inhibitor proteins securin and cyclin B. These studies led to the identification of a phosphate-binding site on cyclin B\, which we showed is an important docking site for mitotic substrates of the cyclin-dependent kinases.

DESCRIPTION:Andrew Stergachis Associate Professor\, Department of Molecular and Medical GeneticsUniversity of Washington \n\n\n\n“The diploid chromatin epigenome: what’s the difference?”Advances in long-read sequencing and genome assembly now enable the routine generation of near-complete diploid telomere-to-telomere (T2T) genomes. As the genetic landscape of these diploid T2T genomes comes into focus\, the next major challenge is to understand the functional differences between the two haploid genomes that comprise an individual’s diploid genome. I will present recent advances from our team in long-read single-molecule epigenetic mapping that enable resolution of the complete diploid chromatin epigenome at both the single-molecule (Fiber-seq) and single-cell (DAF-seq) levels. Using data generated through the Somatic Mosaicism across Human Tissues (SMaHT) Network\, the Human Pangenome Reference Consortium (HPRC)\, the Undiagnosed Diseases Network (UDN)\, and the GREGoR Consortium\, I will demonstrate how germline genetic variation\, somatic epimutations\, and stochastic gene regulatory processes shape the diploid chromatin epigenome. Collectively\, these studies provide a new framework for measuring and conceptualizing function within the diploid T2T genome.
