DESCRIPTION:Seminar held in person only. \n\n\n\n\nDr. Jessica WhitedAssociate Professor\, Department of Stem Cell and Regenerative BiologyHarvard University \n\n\n\n“Systemic controls on axolotl limb regeneration” \n\n\n\nUnderstanding how injury provokes cells to mount the correct response to optimize regeneration is critical for future therapeutics stimulating human limb regeneration. Comparative approaches across species promise to bridge the gap between “super-regenerators” and humans\, but we first must tease out the principles operating in these super-regenerative species. Axolotls grow a blastema\, a transient structure formed at the injury site\, to coalesce activated progenitor cells to create new tissues during limb regeneration. We are studying the mechanisms whereby blastema cells are specified and the extent to which they act systemically\, versus locally\, in this process. I will highlight our recent work on how adrenergic signaling systemically primes cells toward regeneration\, while also promoting local blastema cell specification at the injury site. I will also share our ongoing work investigating the role of pituitary-derived prolactin in limb regeneration.

DESCRIPTION:Seminar held in person only. \n\n\n\n\nDr. Sara BrownellPresident’s Professor\, School of Life SciencesArizona State University \n\n\n\n“Disclosure decisions: Exploring the concealable stigmatized identities of science instructors and how revealing can impact undergraduates” \n\n\n\nHaving instructors as role models in the undergraduate science classroom has been shown to be important\, yet little is known about the representation of instructor identities that are not visibly apparent. Concealable stigmatized identities are identities that have some negative stereotype and are hidden\, making it necessary for someone to actively reveal these identities. We emailed every science faculty member in the United States at research institutions and were able to document the prevalence of concealable stigmatized identities\, the extent to which instructors revealed these identities to students\, and the representation gaps between instructors and students with these identities. Further\, we explored factors that contributed to instructors either revealing or concealing these identities. Few instructors reveal concealable stigmatized identities to students\, yet we established that when instructors revealed these identities to students in class\, all students benefit and students with that specific concealable stigmatized identity benefit disproportionately. This work highlights the potential impact that instructors with concealable stigmatized identities can make and pinpoints the need to consider concealable stigmatized identities in our effort to promote diversity and inclusion.

DESCRIPTION:Seminar held in person only. \n\n\n\n\nDr. Shu-Hsien SheuGroup LeaderBiohub \n\n\n\n“A serotonergic axon-cilium synapses” \n\n\n\nChemical synapses are classically formed between axons and dendrites — yet we discovered that serotonergic axons also synapse directly onto neuronal primary cilia\, organelles positioned at the nexus of the cell surface and the nucleus. Understanding what signals flow through this synapse\, and how it operates in human neurons\, is the focus of our work.We previously showed\, using enhanced focused ion beam–scanning electron microscopy (FIB-SEM) with optimally preserved ultrastructure\, that brainstem serotonergic axons form synapses with the primary cilia of hippocampal CA1 pyramidal neurons — the axo-ciliary synapse. These cilia are selectively enriched in the ciliary-restricted serotonin receptor 5-HTR6\, and optogenetic and chemogenetic stimulation of serotonergic axons drives serotonin release directly onto cilia. Ciliary 5-HTR6 activation engages a non-canonical Gαq/11–RhoA pathway that remodels nuclear actin and increases histone acetylation and chromatin accessibility\, revealing a more direct route from neuromodulatory input to epigenetic state.Building on this foundation\, my group is now establishing the human biology of neuronal primary cilia and axo-ciliary synapses. We are developing human induced pluripotent stem cell derived neuronal models and quantitative imaging approaches\, including fluorescence lifetime imaging\, to probe ciliary second messenger signaling\, alongside cryo–electron tomography to resolve their molecular architecture. In parallel\, we are building high-throughput in vitro platforms to reconstitute axo-ciliary synapses\, laying the groundwork for systematic interrogation of this synapse class in a human context.
